2-Oxoisovalerate palmitoylcarnitine and 2-oxoisovalerate were 1 .O, 0.3 and 0.07 respectively. The rate of oxidation of 2-oxoisovalerate determined spectrophotometrically was about 12 times faster than when measured at 30°C by "CO, release from 2-0xo[ 114C]isovalerate by rat liver mitochondria1 fractions (Bremer & Davis, 1978; Patel et al., 1980). a-Cyano-4-hydroxycinnamate specifically inhibits pyruvate uptake by mitochondria (Halestrap et al., 1980). There was no inhibition of the oxidation of 0.5 mM-2-OXOiSOValerate by 100 j~-a-cyano-4-hydroxycinnamate nor by 10 mM-pyruvate under our experimental conditions, in apparent disagreement with other reports (Halestrap et al., 1980; Patel et al., 1980). This leaves unanswered the question of the nature of the carriers by which 2-oxoisovalerate enters the mitochondrial matrix. Direct spectrophotometric measurements of branched-chain 2-0x0 acid dehydrogenase with 2-oxoisovalerate as substrate will be useful to study the activity of this enzyme in liver in different hormonal and nutritional conditions.

Skin fibroblasts obtained from cancer patients display foetal-like migratory behaviour on collagen gels.
When plated on the surface of collagen gel substrata, all types of fibroblasts rapidly begin to migrate down into the three-dimensional collagen matrix. We have previously demonstrated that normal (adult and foreskin), foetal and transformed fibroblasts may be distinguished from each other by virtue of their differential migratory response to changes in cell density. The effects of cell density...
متن کاملDetection of the Duplication in Exons 56-63 of Duchenne Muscular Dystrophy Patients with MLPA
Background Duchenne Muscular Dystrophy (DMD) is a deadly X-linked recessive disorder. This genetic disorder affects 1 among 3,500-5,000 males in the world. The majority of the patients are male, due to the type of inheritance. It affects most of the skeletal, the respiratory, and cardiac muscles, causing these vital organs to contract and eventually mortality.<br...
متن کاملSpreading behaviour of cultured fibroblasts from carriers of Duchenne muscular dystrophy.
Cultured skin fibroblasts from patients with Duchenne muscular dystrophy (DMD) are more sensitive than normal cells to prolonged exposure to the ionophore monensin. In a cell spreading assay in which cells were preincubated with monensin and subsequently allowed to adhere to and spread on a glass substratum in serum-free medium for 100 min, the mean transformed cell area of normal and DMD cells...
متن کاملThe kinetics of cell-substratum detachment mediated by trypsin: a comparison of normal and Duchenne fibroblasts.
In previous studies of cell-cell and cell-substratum adhesion, we have identified differences in the behaviour between human skin fibroblasts cultured from normal individuals and patients with Duchenne muscular dystrophy (DMD). In these studies, monolayer cultures were dissociated by trypsinization and no detectable difference was noted in the efficiency of cell dissociation between normal and ...
متن کاملDecrease in the rate of protein synthesis by polysomes from cultured fibroblasts of patients and carriers with Duchenne muscular dystrophy.
Polysomes extracted from cultured fibroblast cells isolated from patients with Duchenne muscular dystrophy (DMD), carriers of the disease, and normal controls were used for in vitro measurement of protein synthesis in a wheat germ extract system. It was observed that polysomes from patients and carriers (seven of each aged 17 years or older) exhibited a 3-fold and a 1.5-fold decrease in the rat...
